Medicinal Chemistry I (BP402T): Unit 1 - SAR, Physicochemical Properties & Receptors

Compiled by Prof. Dr. N. K. Aggarwal & Team

Chapter 1: Physicochemical Factors & Drug Design

The biological activity of a pharmaceutical compound is fundamentally governed by its physicochemical parameters: Ionization (\(pK_a\)), Lipophilicity (\(\log P\)), Hydrogen bonding, and Steric geometry.

1.1 Hansch Analysis (Quantitative SAR)

\[ \log \left(\frac{1}{C}\right) = -k (\log P)^2 + k'(\log P) + \rho \sigma + \delta E_s + \text{constant} \]

Balances the parabolic lipophilicity model with electronic Hammett parameters (\(\sigma\)) and Taft steric factors (\(E_s\)).
